Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.
Two structural changes distinguish the synthetic peptide from the natural hormone. A norleucine residue replaces methionine at one position, and a D-configured phenylalanine replaces the natural L-form at another. Both substitutions slow enzymatic breakdown, which extends the molecule's persistence relative to the parent hormone. The lactam bridge further constrains the backbone into a stable conformation. These features are standard design strategies in peptide chemistry and are not unique to this compound; they appear across many research peptides built for improved stability.
Scientific discussion of Melanotan-2 spans pharmacology, dermatology, and public-health literature. Laboratory studies examine its receptor binding and cellular effects, while clinical reports describe outcomes observed after unregulated use. These two bodies of work differ in rigour and intent. Peer-reviewed trials of the compound as a medicine are limited, so much of the available information comes from case reports and surveillance data. Authors frequently note the gap between experimental findings and real-world use.
Reported observations after unregulated use include shifts in skin pigmentation and, in some accounts, unintended changes to moles and other lesions. Whether these outcomes are causally linked to the compound, and how often they occur, remain open questions because controlled data are scarce. The absence of standardised dosing and verified product purity complicates interpretation. Researchers have called for better surveillance and analytical characterisation of samples obtained outside regulated channels. Conclusions drawn from anecdotal evidence should be treated as provisional.

Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Irradiation of 241Am by 12C or 22Ne ions yields the isotopes 247Es (einsteinium) or 260Db (dubnium), respectively. Furthermore, the element berkelium (243Bk isotope) had been first intentionally produced and identified by bombarding 241Am with alpha particles, in 1949, by the same Berkeley group, using the same 60-inch cyclotron. Similarly, nobelium was produced at the Joint Institute for Nuclear Research, Dubna, Russia, in 1965 in several reactions, one of which included irradiation of 243Am with 15N ions. Besides, one of the synthesis reactions for lawrencium, discovered by scientists at Berkeley and Dubna, included bombardment of 243Am with 18O.
Trimebutine nitroarginate and derivatives – arginine nitro-derivative acts as a nitric oxide donor, which exerts anti-inflammatory effect (similarly to naproxcinod) Trimebutine thiocarbamoylbenzoate – thioamide group acts as a hydrogen sulfide donor, also acting as an anti-inflammatory and analgesic agent
== Signs and symptoms == Patients with Sack–Barabas syndrome have thin, fragile skin, especially in the chest and abdomen, that bruises easily; hands and feet may have an aged appearance. Skin is soft but not overly stretchy. Facial features are often distinctive, including protruding eyes, a thin nose and lips, sunken cheeks, and a small chin. Other signs of the disorder include hypermobility of joints, tearing of tendons and muscles, painfully swollen veins in the legs, lung collapse, and slow wound healing following injury or surgery. Infants with the condition may be born with hip dislocations and clubfeet. Unpredictable ruptures of arteries and organs are serious complications of SBS. Ruptured arteries can cause internal bleeding, stroke, or shock, the most common cause of death in patients with this disorder. Rupture of the intestine is seen in 25 to 30 percent of affected individuals, and tearing of the uterus during pregnancy affects 2 to 3 percent of affected women. Although these symptoms are rare in childhood, more than 80 percent of patients experience severe complications by the age of 40. Teenage boys are at high risk for arterial rupture, which is often fatal.
=== Cancer === In a 2022 update of an earlier report, the United States Preventive Services Task Force recommended against the use of vitamin E supplements for the prevention of cardiovascular disease or cancer, concluding there was insufficient evidence to assess the balance of benefits and harms, yet also concluding with moderate certainty that there is no net benefit of supplementation. As for literature on different types of cancer, an inverse relationship between dietary vitamin E and kidney cancer and bladder cancer is seen in observational studies. A large clinical trial reported no difference in bladder cancer cases between treatment and placebo. An inverse relationship between dietary vitamin E and lung cancer was reported in observational studies, but a large clinical trial in male tobacco smokers reported no impact on lung cancer between treatment and placebo, and a trial which tracked people who chose to consume a vitamin E dietary supplement reported an increased risk of lung cancer for those consuming more than 215 mg/day. For prostate cancer, there are also conflicting results. A meta-analysis based on serum alpha-tocopherol content reported an inverse correlation in relative risk, but a second meta-analysis of observational studies reported no such relationship. A large clinical trial with male tobacco smokers and reported a 32% decrease in the incidence of prostate cancer, but the SELECT trial of selenium or vitamin E for prostate cancer enrolled men ages 55 or older and reported relative risk 17% higher for the vitamin group.

The association of anti-tobacco smoking research and public health measures with the Nazi leadership may have contributed to the lack of attention paid to these studies. They were also published in German and Dutch. These studies were widely ignored. In 1947 the British Medical Council held a conference to discuss the reason for the rise in lung cancer deaths; unaware of the German studies, they planned and started their own. Five case-control studies published in 1950 by researchers from the US and UK did draw widespread attention. The strongest results were found by "Smoking and carcinoma of the lung. Preliminary report", by Richard Doll and Austin Bradford Hill, and the 1950 Wynder and Graham Study, entitled "Tobacco Smoking as a Possible Etiologic Factor in Bronchiogenic Carcinoma: A Study of Six Hundred and Eighty-Four Proved Cases". These two studies were the largest, and the only ones to carefully exclude ex-smokers from their non-smokers group. The other three studies also reported that, to quote one, "smoking was powerfully implicated in the causation of lung cancer". The Doll and Hill paper reported that "heavy smokers were fifty times as likely as non-smokers to contract lung cancer".
=== 4 May === Explosions were reported in Kyiv and Odesa in another series of Russian air attacks. Ukraine claimed to have destroyed 18 out of the 24 drones launched, plus a reconnaissance drone, with only light damage to property; the drones were marked with messages such as "For Moscow" and "For the Kremlin". According to TASS, Ukrainian drones attacked and set ablaze the Ilsky refinery near Novorossiisk. The fire was extinguished without damage after two hours. Fighting broke out between Russian and Ukrainian delegates to the Parliamentary Assembly of the Black Sea Economic Cooperation in Ankara, Turkey. Ukrainian delegates had earlier waved Ukrainian flags during a speech by a Russian delegate. Later, Oleksandr Marikovski, a Ukrainian MP, physically assaulted a Russian official after the latter seized his flag. Ukraine for the first time intercepted a Russian Kh-47M2 Kinzhal hypersonic missile, following reports from Kyiv of a powerful explosion overnight. The Ukrainian Air Force said they shot it down using the Patriot missile system protecting the capital.
Acute coronary syndrome, e.g., NSTEMI Atrial fibrillation Deep-vein thrombosis and pulmonary embolism (both prevention and treatment) Other thrombotic states and conditions Cardiopulmonary bypass for heart surgery ECMO circuit for extracorporeal life support Hemofiltration Indwelling central or peripheral venous catheters Heparin and its low-molecular-weight derivatives (e.g., enoxaparin, dalteparin, tinzaparin) are effective in preventing deep vein thromboses and pulmonary emboli in people at risk, but no evidence indicates any one is more effective than the other in preventing mortality. In angiography, 2 to 5 units/mL of unfractionated heparin saline flush is used as a locking solution to prevent the clotting of blood in guidewires, sheaths, and catheters, thus preventing thrombus from dislodging from these devices into the circulatory system . Unfractionated heparin is used in hemodialysis. Compared to low-molecular-weight heparin, unfractionated heparin does not have prolonged anticoagulation action after dialysis and is low cost. However, the short duration of action for heparin would require it to maintain continuous infusion to maintain its action. Meanwhile, unfractionated heparin has higher risk of heparin-induced thrombocytopenia.
=== Vaporization === Ethnobotanist Daniel Siebert cautions that inhaling hot air can be irritating and potentially damaging to the lungs. Vapor produced by a heat gun needs to be cooled by running it through a water pipe or cooling chamber before inhalation.

=== Targeting Heme === Heme, an important prosthetic group present in Complexes I, II, and IV can also be targeted, since heme biosynthesis and uptake have been correlated with increased cancer progression. Various molecules can inhibit heme via different mechanisms. For instance, succinylacetone has been shown to decrease heme concentrations by inhibiting δ-aminolevulinic acid in murine erythroleukemia cells. The primary structure of heme-sequestering peptides, such as HSP1 and HSP2, can be modified to downregulate heme concentrations and reduce proliferation of non-small lung cancer cells.

It is a synthetic seven-amino-acid peptide modeled on alpha-melanocyte-stimulating hormone. It carries two non-natural substitutions and a cyclic bridge that increase its stability relative to the natural hormone. It circulates as a research chemical and is not an approved medicine.
Melanotan-1, also called afamelanotide, is a shorter linear analog with a different amino acid sequence and no lactam ring. It has been evaluated in formal clinical programs, while melanotan-2 has not. The two are distinct molecules and are not interchangeable.
No. Bremelanotide is a related but distinct cyclic peptide that reached approved status for one specific clinical indication. Melanotan-2 is a separate molecule with its own sequence and properties. Shared ancestry in the melanocortin family does not make them the same substance.
Major regulatory agencies have not approved it for any indication. Some countries permit it only under prescription frameworks, while others classify it as a controlled substance.
