The short version of melanogenesis fits in a sentence. The long version — which is the one that helps — is below.

Identity and purity are assessed with chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the target peptide from related impurities and degradation products, and the resulting retention time is compared against a reference standard. Mass spectrometry, often coupled to liquid chromatography, confirms molecular mass. Amino acid analysis or peptide mapping can provide additional sequence-level confirmation when required. Results are only as reliable as the reference materials used alongside them.
Regulatory treatment varies by country. In the United States, melanotan-2 is not approved for any indication, and products marketed for human use fall outside the approved drug framework. Some other jurisdictions have placed it under prescription controls or listed it as a prohibited or restricted substance. Online listings frequently describe the material as a research chemical, a category that does not carry the same manufacturing and labelling requirements as approved medicines.
Solid peptide material is generally stable when kept cold and dry. Common practice is storage at -20 degrees Celsius or lower, with desiccant and protection from light. Repeated freeze-thaw cycles and exposure to moisture are associated with degradation, aggregation, or loss of material. Once dissolved, stability depends on solvent, concentration, and temperature, and solutions are usually treated as short-lived unless stability data support longer periods. Handling notes typically emphasise minimising time at ambient temperature.
Human data remain limited and mostly short-term. Reports describe small trials and observational accounts rather than large controlled studies, so questions about dose-response relationships and long-term effects on melanocytes stay open. Whether repeated exposure alters naevus behaviour is not settled in the published record. Researchers also note that self-administered use outside clinical settings makes actual exposure difficult to quantify. Statements about efficacy and safety should therefore be read as preliminary rather than established.
Melanotan II is a synthetic cyclic heptapeptide that acts as an agonist at melanocortin receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide involved in pigment production. The analogue carries a lactam bridge that constrains the ring and slows enzymatic breakdown relative to the native hormone. In research literature it appears under several abbreviations, and naming conventions are not fully standardized. Published descriptions usually place it within the broader melanocortin agonist family.
Receptor binding at MC1R on melanocytes raises intracellular cyclic AMP and increases expression of tyrosinase and related enzymes. The downstream result is greater synthesis of eumelanin, the dark pigment, without ultraviolet exposure acting as the trigger. The compound is not selective, however, and also engages MC3R, MC4R and MC5R, which are expressed in the central nervous system and elsewhere. That lack of selectivity is the explanation usually offered for effects reported outside pigmentation, including appetite suppression and nausea. Selectivity remains a central theme in comparative studies of related peptides.

Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.
Identity testing for a cyclic peptide of this size usually relies on reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The mass spectrum confirms molecular weight, while the chromatographic trace indicates the proportion of related impurities. Tandem mass spectrometry can provide sequence-level information when fragmentation data are compared against a reference standard. Nuclear magnetic resonance is sometimes used to confirm the lactam bridge, although it requires more material and greater operator expertise than routine chromatographic methods.
Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Identification in laboratories relies on reversed-phase liquid chromatography coupled with tandem mass spectrometry, with product-ion spectra compared against a certified reference standard. High-resolution mass spectrometry supplies accurate mass confirmation, and peptide mapping after enzymatic digestion separates melanotan II from closely related analogues. Quantitation of seized material is complicated by unknown counter-ions and residual trifluoroacetate left from purification. Immunoassays raised against alpha-melanocyte-stimulating hormone can cross-react, so chromatographic confirmation is normally required. Urinary detection windows are short, and reported limits of detection differ substantially between laboratories.
Melanotan II holds no marketing authorisation from the Food and Drug Administration, the European Medicines Agency, the UK Medicines and Healthcare products Regulatory Agency or Australia's Therapeutic Goods Administration. Products sold under that name are treated as unapproved new drugs, and their sale or import is prohibited in several jurisdictions. Other countries classify the peptide as a prescription-only medicine or place it among controlled substances, so the legal position changes with the destination market. No pharmacopoeial monograph supplies an official specification, because the material is not a licensed pharmaceutical. Consequently, products offered online are not manufactured to a shared public standard.
== History == Its existence was first suggested in 1905 by the British physiologist John Sydney Edkins, and gastrins were isolated in 1964 by Hilda Tracy and Roderic Alfred Gregory at the University of Liverpool. In 1964 the structure of gastrin was determined.
Common adverse effects (2.4–42%) included agitation or irritability, tachycardia, hallucinations or delusions, confusion, pupil dilation, hypertension, drowsiness or lethargy, elevated creatine phosphokinase (CPK), nausea and vomiting, and others. Selected serious adverse effects included fever or hyperthermia in 3.8%, single seizure in 2.4%, coma in 1.4%, elevated creatinine in 1.4%, multiple seizures in 1.2%, rhabdomyolysis in 1.1%, respiratory depression in 0.9%, cardiac conduction disorder in 0.5%, and status epilepticus in 0.4%. There is a case report of severe neurological sequelae following a single typical recreational dose of LSD involving seizure and cardiorespiratory arrest. In general, psychedelics like LSD may rarely cause seizures in some individuals. The median lethal dose (LD50) of LSD in animals varies and is 50 to 60 mg/kg in mice, 16.5 mg/kg in rats, and 0.3 mg/kg in rabbits all given by injection. A well-known 1962 instance of an elephant named Tusko given 297 mg (~0.1 mg/kg) LSD by intramuscular injection proved fatal. These findings suggest that elephants may be much more sensitive to LSD in overdose than humans and other species. However, this instance has been mired in criticism and controversy due to miscalculation of LSD dose and concomitant post-LSD administration of promazine and pentobarbital. The experiment was repeated in two elephants with similar doses of LSD in 1984 without incident. Massive doses of LSD are largely managed by symptomatic treatments, and agitation can be addressed with benzodiazepines.
=== Cardiac biomarkers === There are two main cardiac biomarkers used in the assessment of cardiac amyloidosis, troponin and N-terminal proBNP. As expected, with cardiac damage and dysfunction, there can be an elevation of these markers in patients with cardiac amyloidosis. These markers have been incorporated into the various staging/scoring systems used by physicians to determine severity of the disease and prognosis.
In June 2000, the 57 countries that have not yet achieved elimination of neonatal tetanus were ranked and the Philippines was listed together with 22 other countries in Class A, a classification for countries close to maternal and neonatal tetanus elimination.

Lesbians are particularly targeted for gang rape. African lesbians are more likely to be raped as lesbians in the townships. To what extent are coloured lesbians also targeted for rape because of their sexual orientation? There are no statistics for this, and I don't know what percent of coloured lesbians are targeted for corrective rape action. Growing up, I never heard that lesbians were targeted in this way and so I want to know when that started happening. Gangsterism has always existed in the townships, so you can't attribute it to that. I don't know why black lesbians are targeted more, either. I'd like to know how many women are being raped by brothers, fathers, etc., in coloured townships. Why is no one studying this? Has it just been under-reported, not studied, or what? The United Nations UNAIDS 2015 Terminology Guidelines suggests that the term corrective rape should no longer be used, as it gives off the perception that something needs to be fixed. The guidelines propose that the term homophobic rape should be used instead. The words "so-called 'curative' or 'corrective' rape" were mentioned in 2011 in the first UN report on discrimination and violence against LGBTQ+ people. A 2013 global study on HIV/AIDS suggested the term lesphobic rape to emphasize the fact that lesbians constitute the overwhelming majority of victims of this phenomenon. Others have emphasized that other members of the LGBTQ community can also be victims of corrective rape for similar reasons.
primary structure The first order of complexity of structural organization exhibited by a polypeptide or protein molecule, or by a polynucleotide or nucleic acid molecule; i.e. the specific linear sequence of amino acid residues, or of nucleotides, without regard to their spatial arrangement or conformation or any higher-order structure (either secondary, tertiary, or quartenary) the molecule might assume. By convention, the primary structure of a polypeptide is reported from the amino (N)-terminus to the carboxyl (C)-terminus, and the primary structure of a nucleic acid is reported from the 5'-end to the 3'-end.
). This diffusivity leads to a break in the magnetic topology; magnetic field lines can "reconnect" when they collide. Usually, this term is small, and reconnections can be treated as similar to shocks; this process has been shown to be important in Earth-solar magnetic interactions. Extended Extended MHD describes a class of phenomena in plasmas that are higher-order than resistive MHD, but can adequately be treated with a single-fluid description. These include the effects of Hall physics, electron pressure gradients, finite Larmor radii in the particle gyromotion, and electron inertia. Two-fluid Two-fluid MHD describes plasmas that include a non-negligible Hall electric field. As a result, the electron and ion momenta must be treated separately. This description is more closely tied to Maxwell's equations because an evolution equation for the electric field exists. Hall In 1960, M. J. Lighthill criticized the applicability of ideal or resistive MHD theory for plasmas. This concerned the neglect of the "Hall current term" in Ohm's law, a frequent simplification made in magnetic fusion theory. Hall magnetohydrodynamics (HMHD) takes into account this electric-field description of magnetohydrodynamics, and Ohm's law takes the form:
=== Other functions === EF-Tu has been found in large quantities in the cytoskeletons of bacteria, co-localizing underneath the cell membrane with MreB, a cytoskeletal element that maintains cell shape. Defects in EF-Tu have been shown to result in defects in bacterial morphology. Additionally, EF-Tu has displayed some chaperone-like characteristics, with some experimental evidence suggesting that it promotes the refolding of a number of denatured proteins in vitro. EF-Tu has been found to moonlight on the cell surface of the pathogenic bacteria Staphylococcus aureus, Mycoplasma pneumoniae, and Mycoplasma hyopneumoniae, where EF-Tu is processed and can bind to a range of host molecules. In Bacillus cereus, EF-Tu also moonlights on the surface, where it acts as an environmental sensor and binds to substance P.
=== Before World War II === The realization that early industrializers like the United States could provide technical assistance to other countries' development efforts spread gradually in the late 1800s, leading to a substantial number of visits to other countries by U.S. technical experts, generally with official support by the U.S. government even when the missions were unofficial. Japan, China, Turkey, and several Latin American countries requested missions on subjects like fiscal management, monetary institutions, election management, mining, schooling, roads, flood control, and urban sanitation. The U.S. government also initiated missions, particularly to Central America and the Caribbean, when it felt that U.S. interests might be affected by crises like failed elections, debt defaults, or spread of infectious disease. U.S. technical missions in this era were not part of a systematic, government-supported program. Possibly the closest approximation to what U.S. government development assistance would become was the China Foundation for the Promotion of Education and Culture, established by the United States in 1924 using funds provided by China as reparations following the Boxer conflict. The foundation's activities ranged widely and included support for development of a leading Chinese university, Tsinghua University.

==== Automation ==== An added benefit of using magnetic beads is that automated immunoprecipitation devices are becoming more readily available. These devices not only reduce the amount of work and time to perform an IP, but they can also be used for high-throughput applications.
If a complexing agent is present in the aqueous phase then it can lower the distribution ratio. For instance, in the case of iodine being distributed between water and an inert organic solvent such as carbon tetrachloride then the presence of iodide in the aqueous phase can alter the extraction chemistry: instead of
The tidal chain reaction theory has mechanistic advantages over thermal association/dissociation at deep-sea vents because it requires that chain assembly (template-driven polymerization) takes place during the dry-down phase, when precursors are most concentrated, whereas thermal cycling needs polymerization to take place during the cold phase, when the rate of chain assembly is lowest and precursors are likely to be more dilute.
== Ion-exchange chromatography == DEAE-C is a weak anion exchanger. This exchange is utilized to separate proteins that have faintly differing charges. Like all anion exchangers, the resin carries a positive charge that interacts favorably with negative charges. The positive charge of DEAE cellulose is due to a protonated amine group. To ensure that the resin is protonated and positively charged, the chromatography should be performed at least 2 pH units below the pKa of the amine group, 10. The strength of the bond between the resin and protein is highly dependent on the pH range in the column and the pI of the protein of interest. The resin is a weak exchanger because it is only partially ionized over most pH values, and an efficient separation with DEAE-C chromatography requires a specific, narrow pH range. Cellulose, dextran, agarose, and other insoluble complexes are unaffected because they compose inert matrices, hence why they are so often derivatized with strong and weak cation and anion exchangers in chromatography. DEAE-C beads have diethylaminoethyl chains covalently bound to oxygen atoms on the D-glucose subunits of cellulose.

Laboratory confirmation typically combines retention time matching on a chromatographic system with mass measurement. A reference standard of known identity is needed for a meaningful comparison. Sequence-level techniques can add further confirmation.
There is no single harmonised standard governing these products, and manufacturing conditions differ. Impurities may include truncated sequences, oxidation products, residual solvents, and counter-ions. Independent testing is often the only way to characterise a given lot.
Reported studies have looked at pigmentation, photoprotection, and metabolic or appetite-related endpoints in small experimental settings. Much of the evidence base is early-stage and limited in size. Questions about long-term effects and clinical relevance remain open.
It is a synthetic cyclic heptapeptide and a non-selective melanocortin receptor agonist. Structurally it is modelled on alpha-melanocyte-stimulating hormone, a peptide that occurs naturally in the body.
